If I ask you which bike would you like to buy in a 125cc segment, what comes in your mind first? Yes, thinking of a 125 cc bike would be mostly related to lower segment bikes that are good for commuting purposes only and you can hardly think of a 125cc bike that would satisfy your needs of features, power, mileage, performance and yes, most importantly, the cool looks that promise to fetch second look from people around.

I am going to list here, Top 5 best looking, hot on the line 125 cc bikes present in the world that money can buy for you.

1. Cagiva Mito 125

Cagiva is a 125cc Italian sports motorcycle. The engine consists of a two-stroke 125 cc single cylinder, with some models capable of producing over 30 bhp. Not many people know that the Cagiva Mito was the first bike of Valentino Rossi.

2. Aprilia RS 125


The Aprilia RS125 is a GP derived replica sport production motorcycle. It is powered by a single cylinder 2-stroke, Nikasil coated aluminium cylinder block, liquid cooled 124.8 cc Rotax engine. Aprilia has had many Grand Prix victories in the 125 cc and 250 cc 2-stroke classes. This racing experience has been used by Aprilia to produce updated and improved versions of the RS125.

3. Yamaha YZF R125


The Yamaha YZF-R125, introduced in 2008, is a sport bike manufactured by Yamaha Motor Company. The 125 cc engine produces 11 kW (15 hp), making it learner-legal entry level in many countries. The Derbi GPR 125 is a street motorcycle sold by Derbi – Nacional Motor since 2005. The original GPR125 was powered by a single cylinder two-stroke engine with a steel lined cast iron cylinder manufactured by Yamaha, but this bike has now been superseded by an all-new DOHC four-stroke four valve model.

5. Honda CBR 125


The Honda CBR125R is a sport bike fitted with a 125 cc four-stroke engine. The CBR125R first appeared on the market in 2004. It is manufactured in Thailand alongside the CBR150R, the latter for the Far East market.

Fusion of a protein with a tag such as GFP or EYFP can cause the protein to loose its function why is this?

lab report due... I really appreciate the help! I am stuck on these...

other questions...

1. How can you tell if a protein is functional if it is tagged with EYFP? Is there a website that has a list maybe?
2. I did a western blot with Hepa-1 cells tagged with EYFP on the mito, golgi, actin, and tubulin. All of the lanes had one band except for the mito.. why is that? (I really have no idea about this one)
3.Why would the levels of expression of the different EYFP chimeras would be different when expressed in the same cell line? Would you expect the same type of result if proteins were expressed in other cell lines? (such as A7 smooth muscle cells)

To make a cell express GFP, its DNA sequence must be inserted into the cell's own. However, it is difficult (improbable) that you will be able to insert the gene in between 2 genes. You are more likely to stick in GFP code inside some gene.

If you are using GFP to determine if a gene is turned on...aka being transcribed to mRNA and ultimately translated, then the GFP is piggybacking off of the transcription factors of that gene being examined. By inserting GFP into the sequence of the correctly functioning gene, you are now making the sequence of the gene in question, incorrect. The transcription machinery will never be able to produce the same protein from that gene sequence.

Depending on where the GFP is inserted, say far downstream in the gene sequence, perhaps you could have one functioning domain of the protein, but never the full protein.
